Effect of graded normobaric hypoxia on Genome-wide expression in healthy individuals

(Submitter supplied) The regulation of oxygen homeostasis is critical in physiology and disease pathogenesis. Studying transcriptomic profiles after hypoxia exposure could potentially uncover valuable biomarkers for predicting responses. To this end, 6 healthy individuals were recruited and exposed to graded normobaric hypoxia (FiO2 ~ 0.19 up till 0.13 in steps of 2% decrement in 10 minutes). A blood sample was collected before (at room air) and after the graded normobaric hypoxia exposure (at FiO2 ~ 0.13) for the gene-expression profiles. more...

Spatial analysis of microRNA regulation at defined tumor hypoxia levels reveals biological traits of aggressive prostate cancer

(Submitter supplied) Mechanisms regulating the gene expression program at different hypoxia severity levels in patient tumors are not understood. We aimed to determine microRNA (miRNA) regulation of this program at defined hypoxia levels from moderate to severe in prostate cancer. Biopsies from 95 patients were used, where 83 patients received the hypoxia marker pimonidazole before prostatectomy. Forty hypoxia levels were extracted from pimonidazole-stained histological sections and correlated with miRNA and gene expression profiles determined by RNA-sequencing and Illumina bead arrays. more...

RPTOR, a novel target of miR-155, regulates the fibrotic phenotype of cystic fibrosis lung epithelium by upregulating CTGF

(Submitter supplied) Cystic fibrosis (CF) is caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene, the most frequent of which is F508del-CFTR. CF is characterized by excessive secretion of pro-inflammatory chemokines into the airway lumen, inducing a highly inflammatory cellular phenotype. This process triggers fibrosis, causing airway destruction and leading to high morbidity and mortality. more...

Cell-Cell communication between skinkeratinocytes and fibroblats is non-redundantly coded through inflammatory cytokines

(Submitter supplied) Cell-cell communication between keratinocytes and fibroblasts is essential for skin homeostasis, regulation, and regeneration. How extracellular cytokine signaling, intracellular protein signaling and transcriptional regulation initiate and maintain this communication is still largely unknown. Here, we study in vitro the secretome and transcriptome dynamics that establish double paracrine cell-cell communication between normal human keratinocytes (NHK) and human dermal fibroblasts (HDF). more...

GPR158 Activates Cellular Stress Responses in Trabecular Meshwork Cells of the Eye’s Aqueous Outflow Pathways: Implications for Ocular Hypertension

(Submitter supplied) Background: the major risk factor for glaucoma is ocular hypertension, a disorder caused by reduced outflow of aqueous humor through the trabecular meshwork. In a previous pharmacogenomic screen for genes associated with ocular hypertension, we identified the novel G protein-coupled receptor, GPR158, and showed it protects against age-related ocular hypertension in mice. Here we show that the glucocorticoid, dexamethasone, increases the level of accumulated GPR158 protein in the trabecular meshwork of the human eye, ex vivo. more...
